The nanomechanical signature of breast cancer
(Nanowerk News) The spread of cancer cells from primary tumors to other parts of the body remains the leading cause of cancer-related deaths. The research groups of Roderick Lim and Cora-Ann Schoenenberger from the Biozentrum of the University of Basel, reveal in the journal Nature Nanotechnology ("The nanomechanical signature of breast cancer") how the unique nanomechanical properties of breast cancer cells are fundamental to the process of metastasis. The discovery of specific breast cancer ?fingerprints? was made using breakthrough nanotechnology known as ARTIDIS. Lim?s team has now been awarded about 1.2 million Swiss francs from the Commission for Technology and Innovation (CTI) to further develop ARTIDIS.
Breast cancer is the most common form of cancer in women with 5500 patients being diagnosed with the disease in Switzerland each year. Despite major scientific advancements in our understanding of the disease, breast cancer diagnostics remains slow and subjective. Here, the real danger lies in the lack of knowing whether metastasis, the spread of cancer, has already occurred. Nevertheless, important clues may be hidden in how metastasis is linked to specific structural alterations in both cancer cells and the surrounding extracellular matrix. This forms the motivation behind ARTIDIS (?Automated and Reliable Tissue Diagnostics?), which was conceived by Dr. med. Marko Loparic, Dr. Marija Plodinec and Prof. Roderick Lim to measure the local nanomechanical properties of tissue biopsies.

?Fingerprinting? breast tumors
At the heart of ARTIDIS lies an ultra-sharp atomic force microscope tip of several nanometers in size that is used as a local mechanical probe to ?feel? the cells and extracellular structures within a tumor biopsy. In this way, a nanomechanical ?fingerprint? of the tissue is obtained by systematically acquiring tens of thousands of force measurements over an entire biopsy. Subsequent analysis of over one hundred patient biopsies could confirm that the fingerprint of malignant breast tumors is markedly different as compared to healthy tissue and benign tumors. This was validated by histological analyses carried out by clinicians at the University Hospital Basel, which showed a complete agreement with ARTIDIS. Moreover, the same nanomechanical fingerprints were found in animal studies initiated at the Friedrich Miescher Institute.
Plodinec, first author of the study, explains: ?This unique fingerprint reflects the heterogeneous make-up of malignant tissue whereas healthy tissue and benign tumors are more homogenous.? Strikingly, malignant tissue also featured a marked predominance of ?soft? regions that is a characteristic of cancer cells and the altered microenvironment at the tumor core. The significance of these findings lies in reconciling the notion that soft cancer cells can more easily deform and ?squeeze? through their surroundings. Indeed, the presence of the same type of ?soft? phenotype in secondary lung tumors of mice reinforces the close correlation between the physical properties of cancer cells and their metastatic potential.
ARTIDIS in the clinics
?Resolving such basic scientific aspects of cancer further underscores the use of nanomechanical fingerprints as quantitative markers for cancer diagnostics with the potential to prognose metastasis.?, states Loparic, who is project manager for ARTIDIS. On an important practical note, a complete biopsy analysis by ARTIDIS currently takes four hours in comparison to conventional diagnostics, which can take one week. Based on the potential societal impact of ARTIDIS to revolutionize breast cancer diagnostics, Lim?s team and the Swiss company Nanosurf AG have now been awarded about 1.2 million Swiss francs by the Commission for Technology and Innovation (CTI) to further develop ARTIDIS into a state-of-the-art device for disease diagnostics with further applications in nanomedicine.
Over the next two years, Lim and colleagues will engage and work closely with clinicians to develop ARTIDIS into an easy-to-use ?push-button? application to fingerprint diseases across a wide range of biological tissues. As a historical starting point, the first ARTIDIS demo-lab has already been established at the University Hospital Eye Clinic to collect data on retinal diseases with the goal of improving treatment strategies.

Last week the New York Building Congress issued a bullish report on the city?s construction industry. It said that spending would reach $30.7 billion this year, the highest total since 2008, and remain at that level for the next several years.

However, the headline and the reality are somewhat different, it turns out.

First, the increase in dollar value is driven in large part by inflation. While the consumer price index is rising at an annual rate of about 2% a year, cost increases in construction over the past three years have been on the order of 20%, estimates Building Congress President Richard Anderson. In real terms, the report notes, there has been no increase in construction spending in the past three years. Part of this is the rebound in material prices from the lowest of the Great Recession, exacerbated by the increase in the price of oil. Don?t forget that recent contracts provide wage increases, even as they implement work rule changes.

The second?and?most alarming?note is that employment in the industry is expected to decline slightly this year, to 110,800, the lowest number since 1998. The lack of any real boost in activity partly explains the decline.

Productivity gains accounts for the rest, but productivity in building works somewhat different than in other industries, Mr. Andersen says. It isn?t primarily the introduction of technology, which has had only some impact in back-office operations. It is the fact that only the best workers have jobs, and they get things done faster and more efficiently.

Here are the employment numbers since 1998.? The Building Congress sees no uptick in jobs until 2014.

The bottom line: Construction is a source of good, middle-class jobs?whether unionized or non-union. It looks like this sector will not be a source of those middle-class jobs.

Russia Fact Check: Romney 'Foe' or Obama 'Reset'?

MOSCOW - When Mitt Romney steps on the stage tonight in Boca Raton, Fla., to debate foreign policy with President Obama, his challenge will often be to draw a distinction between his proposals and the president's policies. In many areas, notably Iran, their positions are very similar. But one area where the Republican candidate and the president differ sharply is on their view of Russia. During an interview with CNN in March, Romney called Russia the United States' "number one geopolitical foe." But is that a fair characterization, or just campaign trail bluster? Analysts are divided, but most admit there is evidence to support both sides. "Russia is clearly not America's number one geopolitical foe. It lacks the economic and conventional military capability to live up to that mantle," Stephen Flanagan, a Russia expert at the Center for Strategic and International Studies, told ABC News by email. "That said, the Kremlin is undermining stability in the Middle East and the effectiveness of the U.N." "I would say that Gov. Romney's assertion is motivated by a calculation that a strong stance on Russia plays well with a segment of the American electorate. Given the challenges posed by international terrorism, Afghanistan, turmoil in the broader Middle East and a rising China, it is very difficult to see Russia as our number one problem," said Stephen Pifer, a former U.S. Ambassador to Ukraine and now an analyst at the Brookings Institute. "It's political hyperbole to suggest they are our biggest problem," Matthew Rojansky, the deputy director of the Russia and Eurasia Program at the Carnegie Endowment, told ABC News. "There is something to it in the sense that if you look at the world through the prism of the right side of some issues and the wrong side, Russia tends to be on the other side of us. And if you've picked the right side, then Russia is on the wrong side." Others noted that Romney's remarks represent the views of many in his party. "Gov. Romney's statement is reflective of widespread disappointment with the reset among many Republicans. Many see Moscow as on the opposite side from the United States in dealing with major international issues, particularly Syria, and believe this is clear evidence that the reset has failed," said Paul Saunders, the executive director at the Center for the National Interest. Romney's remarks did not go unnoticed in the Kremlin. Then-Russian President Dmitri Medvedev immediately dismissed the comments, saying Romney was stuck in the Cold War. But Romney doubled down in an op-ed on Foreign Policy magazine's website and said President Obama's policies of outreach were to blame. He held his ground in another CNN interview in July, saying: "There's no question but that in terms of geopolitics - I'm talking about votes at the United Nations and actions of a geopolitical nature - Russia is the No. 1 adversary in that regard." Six months later, however, President Vladimir Putin said Romney's comments would only harden Russia's stance, particularly its opposition to U.S.-led plans to place a missile defense system in Eastern Europe to guard against an attack by Iran. Russia has insisted in written assurances to guarantee the system is not aimed at its own nuclear arsenal. The Obama administration, meanwhile, has tried to fight the perception that its Russia policy has failed. Shortly after taking office in 2009, the administration launched what it called a "reset" in relations with Moscow. While that initially yielded what many analysts agree was substantially improved relations with the Kremlin, relations have again soured, in large part due to sharp disagreements on policies in the Middle East. Indeed, in justifying his characterization of Russia in March, Romney pointed out that Russia often stands in the way of key American foreign policy goals. After supporting tougher sanctions on Iran for its defiance of international demands to halt its nuclear enrichment activities, Russia reversed course and blocked subsequent efforts to impose additional sanctions. In Libya in 2011, the Kremlin accused the U.S.-led coalition of misleading Russia into supporting a U.N. intervention that gave cover to a NATO-led military operation that helped oust longtime leader Moammar Gadhafi. And the difference between Moscow and Washington has perhaps most acutely been seen in Syria, where Russia has appeared eager to back the government of President Bashar al-Assad, Russia's last ally in the region and a major arms client. Russia even vetoed United Nations Security Council resolutions that would have imposed sanctions on the Assad government. More recently, Putin provided more evidence that relations have deteriorated. He has accused the United States of funding the unprecedented wave of protests against his government. In the past six weeks Russia has expelled the U.S. Agency for International Development and announced it will suspend cooperation on a highly successful deal to secure loose Soviet era nuclear material. Obama, however, has maintained a willingness to engage Russia. On March 26, he was caught on an open microphone telling Medvedev he would have more "flexibility" to negotiate the missile defense issue after the November election. The Romney campaign immediately pounced on that as a sign of Obama's weakness (and prompting Romney's "foe" comment.) Most analysts, however, say that while Obama's efforts yielded progress like signing New START, which reduces each country's nuclear weapons stockpile, by almost any measure the "reset" is over. "No matter the outcome on Nov. 6, the U.S. will need a comprehensive review of its policy toward Russia," said Heather Conley, the director of the center for Strategic and International Studies' Europe Program. She believes the reset ended with the spat of intervention in Libya. "We are now post-reset," Rojansky agreed.

Yahoo Faces Mayer Dilemma: Stay A Media ... - The Next Web

A drastic turn in Yahoo?s future could be announced tomorrow as the search engine company releases its first quarterly earnings report since it brought on Marissa Mayer as CEO. The former executive at Google has made some moves during her short tenure as the leader, but Reuters is reporting that the next announcement on the revival of the struggling search engine will be on Monday when it reports its quarterly results.

For years, the company has become enamored with content creation, signing deals with ABC News,?CNBC, and DailyMotion?to share content, among others. But with the hiring of Mayer, the company is changing things up in order to restore the company to its former glory. And that means that it?s faced with its most complex decision in recent history: does it abandon media-sharing and content creation in favor of building out a more social product?

Yahoo needs to find something that will bring people back to its network of sites and engage with the content more. Reuters says that?Yahoo has been criticized for allowing these sites to ?stagnate? ? there?s not much appeal to them and nothing will drive people back to it. Its CEO wants to make it more interactive, adaptable, and useful on the myriad of devices out there, including tablets and mobile devices.

To that end, is abandoning its content sharing partnerships entirely the smart move for Yahoo? In its 2011 annual report, Yahoo recognized that its stiffest competition is going to come from Facebook, Microsoft, AOL, and, naturally, Google, all of whom offer a variety of Internet products that rival the search engine. Not only that, but advertising networks are helping to make it difficult for the company to compete for a share of advertiser dollars.

With Mayer?s product-oriented background, Yahoo could find itself going after Facebook and Google to reclaim its honor. The company has acquired a lot of companies over the past 18 years, but has yet to fully integrate them into the fold. This new direction presents a 180-degree shift in direction set forth by Mayer?s predecessors and, as Reuters notes, could pose some risks, specifically abandoning media partnerships that have given the company incredible traffic, something championed by former CEO Ross Levinsohn.

Earlier this year, there was an inkling about Mayer potentially taking the payment from the company?s sale of its Alibaba stake?and using it to go after acquisitions of smaller companies, something that Google has been doing over the years. That plan never happened and the money went back to investors. Nevertheless, the opportunity for Yahoo to move and start acquiring small startups to build out its Yahoo social network is ripe for the taking.

Just making improvements to Flickr to compete with the capability of Facebook and Picasa could be a great start to shift the company to a more social business. Other Yahoo-owned companies that could be folded into a potential social play include IntoNow, Dapper, Upcoming.org, and many others. And with Yahoo suing Facebook alleging patent infringement (and who ultimately settled), this could be?the company firing its opening salvo across the bow of Facebook letting it know it?s here to compete.

But with more advertising and media startups that it acquired over the years such as BlueLithium, Interclick, Associated Content, and others, it?s going to be interesting to see how Yahoo can move past its traditional monetization strategy of being all about content, not to mention search and email.

Living Life As It Unfolds: This One's All Mine

I had the opportunity to use this product all by myself, and I love it! ?I know it probably sounds strange that I love learning about taxes, but since I do our personal taxes every year I was eager for this review so that I'd know without a doubt that I was doing them right and would hopefully get some tips to save even more money in the future.

The course has two enrollment options. ?Both options include a 20-lesson course that comes with a manual which has practice problems and examinations, a grading service, practical case studies, student guidance, reference books, a book on how to build and operate a successful tax practice, a graduation certificate, and postgraduate services. ?The first option includes four years of postgraduate support for $795 for payment in full upon registration. ?There are two additional payment plans under this option that allow for installment payments. ?The total amount paid will be more than if paid all at once. ?With the installment plans, only a certain amount of lessons can be completed before the next payment is made.

The second option includes two years of postgraduate support for $495 paid in full upon registration. ?The other payment plan under this option allows for payment in three installments; six lessons can be completed before each succeeding installment is paid. ?Those currently serving in the military may be eligible for reimbursement up to 75% tuition under DANTES.

I have enjoyed the course so far. ?I just finished lesson six, and I have learned a great deal already. ?The one thing that really excites me is the idea of one day being able to have a small business of my own preparing taxes for other people. ?I don't know if that is what God has in store for me, but this course is giving me the knowledge and the opportunity that I've never had before. ?I feel good every time I sit down to take a test, knowing that I'm one step closer to accomplishing this goal in my life. ?Once I've successfully completed the program, I'll be able to take the IRS Registered Tax Preparer Exam. ?I'll have to let you know in a future blog how that turns out.

The lessons include many examples to clarify the text. ?Real life types of examples are given to explain concepts. ?This is extremely helpful to me. ?Sometimes I read through the idea being taught and don't completely understand it. ?When I read the example, the light bulb goes on in my head. ?Each lesson has practice problems at the end for review before you take the test. ?I got most of the answers right for each of the reviews for the first five lessons, but when I got to lesson six the concepts were a bit tougher for me to grasp. ?I didn't get enough right answers for my liking. ?I wished I had had more problems to practice.

Each test that is taken is to be sent in using the envelopes they provide for each test (you need to supply the stamp), and the teachers with National Tax Training School grade your test and mail it back to you within the week. ?The school was on a break when I sent my first tests in, so it took a couple of weeks to get them back; but I have since received feedback on each test I have sent. ?An answer key is sent back with each test to explain the answers and to tell you where to find the answers in the text.

This course is very professional, easy to understand, and the instructors are available to answer any questions you may have. ?They want to be sure you enjoy the course and are getting everything out of it that you can. ?The correspondence I've had with one of the teachers has been very helpful and friendly. ?

WASHINGTON (AP) -- Attorneys, business owners and retirees were among those who helped President Barack Obama maintain slim fundraising supremacy over Republican challenger Mitt Romney last month, with the president raising $181 million as he entered the last full month in the fight to keep his job.

An Associated Press review of newly released financial reports found at least $11 million from lawyers and at least $3 million from investors and bankers, some of whom cooled to the president earlier in this election campaign when critics say he cracked down on Wall Street and pushed for consumer-protection reforms. About $22 million more came from retired Americans, an important bloc of voters likely more tuned in to health care reform and changes in retirement benefits.

Their contributions, among hundreds of thousands, went hand-in-hand with record donations to an outside political group helping Obama win a second term. Still, the president began October with less available money to spend than Romney, as Obama's campaign, the Democratic Party and related groups reported $149.8 million cash on hand. That's compared with $183.1 million in the bank among Romney's campaign, the GOP and his joint-fundraising apparatus.

All told, a swath of small-dollar contributions helped Obama and the Democratic Party best Romney and the GOP by more than $10 million last month after being repeatedly walloped in the money race earlier this year. The president's fundraising haul topped the more than $114 million he and the Democrats raised during the month of August, and the cash Obama pulled in last month was slightly less than his record-breaking $190 million from September 2008.

Financial support to Obama and Romney are putting the presidential election on track to cost nearly $2 billion, thanks to mountains of cash earmarked to both campaigns and independent "super" political committees working on their behalf. Wealthy Americans are increasingly picking up the tab this year, at times giving millions of dollars apiece to super PACs that have buoyed costly advertising.

September's reports show major financial support going to both Republican and Democratic super PACs, with the pro-Romney Restore Our Future PAC reporting it collected $14.8 million in September, the group's second-most lucrative month. Late Saturday, American Crossroads, the major super PAC headed by GOP strategist Karl Rove, reported $11.6 million for September, the highest single-month total for the group.

Rove's top donors in September included Texas billionaire Harold Simmons, who gave $2.5 million and Oklahoma coal baron Joseph Craft III, who gave $1 million. Another $1.32 million was donated by the Armstrong Company, a Pennsylvania conglomerate which provided the donation as "in-kind cable access, and $1 million from Jay Bergman, an Illinois oilman.

Meanwhile, Priorities USA Action, the pre-eminent Obama-supportive super PAC, said it raised a record $15.3 million.

"People who support the president know that we've come too far to go back now," Priorities senior strategist Bill Burton said. "We're ahead a little bit now, but it is time to close the deal."

Both campaigns and super PACs have made an all-out push for contributions as Election Day quickly approaches. The contributions are funding a record-breaking campaign operation for both candidates, which translates to paying a legion of campaign staff and hundreds of millions of dollars' worth of television ads.

So when it came to contributions directly to Obama's campaign, business owners and some Wall Street types helped pick up the tab, even if they didn't make up a majority of total contributions. Obama's donors in September included more than 4,000 CEOs, records show, and his campaign continued to tally millions pouring in from key battleground states.

Yet Obama hardly has a lock on winning the financial fight.

Republican super PACs have helped to match or exceed Obama's TV ad spending in dozens of media markets in battleground states. Ad spending data obtained by the AP from April through early October found pro-Romney spending has exceeded pro-Obama ad spending by at least $65 million across the nine states expected to decide the election: Colorado, Florida, Nevada, New Hampshire, North Carolina, Ohio, Virginia, Iowa and Wisconsin.

Obama broke presidential fundraising records four years ago, but has found himself financially outgunned for much of the summer, thanks in part to super PACs supporting Romney. Meanwhile, an AP review of campaign data this fall found Obama out-raising Romney in most of the 11 states that at one point were pivotal to win the election.

Reports detailing revenues and expenses for the first half of October are due to the Federal Election Commission by Oct. 25. Those will provide the public with the last financial snapshot before the Nov. 6 election.
