Protein that sends 'painful touch' signals identified

ScienceDaily (Feb. 19, 2012) ? In two landmark papers in the journal Nature this week, scientists at The Scripps Research Institute report that they have identified a class of proteins that detect "painful touch."

Scientists have known that sensory nerves in our skin detect pressure, pain, heat, cold, and other stimuli using specialized "ion channel" proteins in their outer membranes. They have only just begun, however, to identify and characterize the specific proteins involved in each of these sensory pathways. The new work provides evidence that a family of sensory nerve proteins known as piezo proteins are ion channel proteins essential to the sensation of painful touch.

The experiments in the new study were conducted in fruit flies, a model system for the sensory nervous system of mammals, where piezo proteins are also expressed, as well as in certain cell types in the ear, kidney, heart, and other tissues. Future studies will focus on the roles of piezo proteins in sensing sound, blood pressure, and related stimuli that press and/or stretch cell membranes.

"Researchers in this field have been trying for decades to identify pressure-transducing ion channel proteins that exist in mammals, and these piezo proteins are exceptionally strong candidates," said Ardem Patapoutian, a professor in the Department of Cell Biology and the Dorris Neuroscience Center at Scripps Research, and a senior investigator for both papers. "We now have solid clues that we can follow up to learn how the mechanotransduction pathway works and how it is disrupted in diseases."

The two papers appear online in Nature on February 19, 2012.

Following the Path of Clues

Patapoutian's laboratory specializes in the study of sensory ion-channel proteins. When hit by a stimulus to which it is sensitive, one of these proteins typically will open its structure to allow charged calcium, sodium, or potassium molecules ("ions") to flow from the fluid outside the cell into the cell's interior. Ion channels that sense mechanical pressure are thought to open when the membrane in which they are embedded is distorted past a certain threshold. The resulting flow of charge can trigger other signals inside the cell, for example a nerve impulse within sensory neurons -- and in a human, a sufficient number of these nerve impulses would be interpreted by the brain as a touch- or pressure-related feeling.

In a highly cited paper published in Science in late 2010, Patapoutian and his colleagues reported that two mouse proteins of previously unknown function exhibited properties of mechanotransducers. Cells to which these proteins were added drew in positively charged ions when subjected to mechanical pressure. Bertrand Coste, the first author of the paper, named the two closely related proteins piezo1 and piezo2 -- the prefix "piezo-" being derived from the ancient Greek word for pressure or squeezing.

"Since these proteins bore little resemblance to known ion channel proteins, the next step for us was to confirm that they are indeed ion channel proteins," Patapoutian said. The new studies take this step and more.

In the first of the new studies, lead authors Bertrand Coste, Bailong Xiao, and their colleagues confirmed that piezo proteins are indeed ion channel proteins, and very large ones. "It assembles into a 'tetramer' complex of four piezo proteins, which appears to be the biggest plasma membrane ion channel yet discovered," said Coste, a research associate in the Patapoutian lab. The protein sequences within piezo also suggest that its ion channel structure weaves through the cell membrane more than 100 times.

Collaborating researchers in the laboratory of Mauricio Montal, a Distinguished Professor of Neurobiology at the University of California, San Diego, found that even in the absence of other proteins, piezo proteins could self-assemble into this tetramer complex, forming ion channels in artificial membranes known as lipid bilayers.

The second of the new studies involved experiments with the fruit fly Drosophila. Sung Eun Kim, first author of the study, genetically engineered a line of Drosophila that does not express the Drosophila piezo (dpiezo) gene. "We found that their larvae showed a severe loss of responsiveness to mechanical stimuli that would be expected to generate pain-related signals, though they responded normally to other kinds of stimuli such as heat and mild pressure," she said. Kim is a graduate student who divides her time between the Patapoutian lab and the lab of Scripps Research Assistant Professor Boaz Cook, who was co-principal investigator of this study.

Kim also used genetic "knockdown" techniques in Drosophila to show that interrupting dpiezo expression in certain sensory neurons could reproduce this loss of sensitivity. Finally, when she artificially reinstated dpiezo expression in larvae that had been born without the gene, they displayed normal sensitivity to strong pressure. "It's the first demonstration of a specific physiological function of a piezo family protein," said Cook.

The Patapoutian lab now is now conducting detailed follow-up studies of piezo and other possible mechanotransduction proteins. "In the next several years, we'll be trying to determine all the biological processes and diseases in which these pressure-sensing proteins play a role," he said.

Other contributors to the first paper, "Piezos Are Pore-Forming Subunits of Mechanically Activated Channels," were J?rg Grandl, Kathryn S. Spencer, Sung Eun Kim, Manuela Schmidt, and Adrienne E. Dubin, all of the Patapoutian lab; Jose S. Santos and Ruhma Syeda of Dr. Montal's lab at UC San Diego; and Jayanti Mathur of the Genomic Institute of the Novartis Research Foundation (where Patapoutian is director of discovery research).

The study was supported by grants from the National Institute of Dental and Craniofacial Research, the National Institute of Neurological Disorders, the National Institute of General Medical Sciences, and by The Genomics Institute of the Novartis Research Foundation. The postdoctoral fellowships of Bailong Xiao and J?rg Grandl are funded by the American Heart Association and the National Institutes of Health, respectively.

Other contributors to the second paper, "The Role of Drosophila Piezo in Mechanical Nociception," were Bertrand Coste of the Patapoutian lab and Abhishek Chadha of the Cook lab. This study was funded by the National Institutes of Health and the Novartis Research Foundation. Kim and Chadha are funded by Scripps Research's Skaggs Institute for Chemical Biology.

(AP) ? Clapping hands and swaying to gospel hymns in the church where Whitney Houston's powerful voice once wowed her congregation, the biggest names in entertainment sang along with the choir to remember the pop superstar at her hometown funeral Saturday.

"We are here today, hearts broken but yet with God's strength we celebrate the life of Whitney Houston," the Rev. Joe A. Carter told the packed New Hope Baptist Church after the choir behind him sang "The Lord is My Shepherd."

Mourners including singer Jennifer Hudson and Houston's mother, gospel singer Cissy Houston, stood, swayed and clapped along in the aisles as gospel singers BeBe Winans and the Rev. Kim Burrell joined with pop stars like Alicia Keys in paying tribute to the 48-year-old pop superstar who first began singing in the Newark church.

"You wait for a voice like that for a lifetime," said music mogul Clive Davis, who shepherded Houston's career for decades.

Others were more mournful; singer Ray J., who spent time with Houston during her last days, broke down crying. His sister, singer Brandy, put her arm around him. Cissy Houston and Houston's daughter, 18-year-old Bobbi Kristina, clutched each other in the front of the row.

Actor Kevin Costner, her co-star in "The Bodyguard" that spawned her greatest hit, remembered a movie star who was uncertain of her own fame, who "still wondered, 'Am I good enough? Am I pretty enough? Will they like me?'"

"It was the burden that made her great and the part that caused her to stumble in the end," Costner said.

Filmmaker Tyler Perry praised Houston's "grace that kept on carrying her all the way through, the same grace led her all the way to the top of the charts. She sang for presidents."

Stevie Wonder and Oprah Winfrey were among the biggest names gathered to mourn Houston, along with Hudson, Monica, Brandy and Jordin Sparks ? representing a generation of big-voiced young singers who grew up emulating her. Houston's voice, a recording of "I Will Always Love You," was to close the funeral.

Houston's cousin Dionne Warwick presided over the funeral, introducing speakers and singers and offering short comments about Houston between them.

Houston's mother was helped by two people on either side of her as she walked in and sat with her granddaughter and other family to begin the service. Houston's ex-husband, Bobby Brown, briefly appeared at her funeral, walking to the casket, touching it and walking out. Security guards said Brown was upset that he would have to sit separately from the people he arrived with, and left. A Brown representative didn't immediately comment.

Mourners fell quiet as three police officers escorted Houston's casket, draped with white roses and purple lilies. White-robed choir members began to fill the pews on the podium. As the band played softly, the choir sang in a hushed voice, "Whitney, Whitney, Whitney."

Close family friend Aretha Franklin, whom Houston lovingly called "Aunt Ree," had been expected to sing at the service, but she was too ill to attend. Franklin said in an email to The Associated Press that she had been up most of the night with leg spasms and sent best wishes to the family. "May God bless and keep them all," she wrote.

A program featuring a picture of Houston looking skyward read "Celebrating the life of Whitney Elizabeth Houston, a child of God." Pictures of Houston as a baby, with her mother and daughter filled the program.

"I never told you that when you were born, the Holy Spirit told me that you would not be with me long," Cissy Houston wrote her daughter in a letter published in the program. "And I thank God for the beautiful flower he allowed me to raise and cherish for 48 years."

"Rest, my baby girl in peace," the letter ends, signed "mommie."

The service marks one week after Houston, one of music's all-time biggest stars, was found dead in a Beverly Hills hotel in California. A cause of death has yet to be determined.

To the world, Houston was the pop queen with the perfect voice, the dazzling diva with regal beauty, a troubled superstar suffering from addiction and, finally, another victim of the dark side of fame.

To her family and friends, she was just "Nippy." A nickname given to Houston when she was a child, it stuck with her through adulthood and, later, would become the name of one of her companies. To them, she was a sister, a friend, a daughter, and a mother.

"She always had the edge," the Rev. Jesse Jackson said outside church Saturday. "You can tell when some kids have what we call a special anointing. Aretha had that when she was 14. ... Whitney cultivated that and took it to a very high level."

A few fans gathered Saturday morning hours before the service as close as they could get to the church, some from as far away as Washington, D.C., and Miami. Bobby Brooks said he came from Washington "just to be among the rest of the fans."

"Just to celebrate her life, not just her death," said Brooks, "just to sing and dance with the people that love her."

Others were more entrepreneurial, setting up card tables to sell silk-screened T-shirts with Houston's image and her CDs. But only the invited would get close to the church; streets were closed to the public for blocks in every direction. But their presence was felt around the church, with a huge shrine of heart-shaped balloons and personal messages that covered the street corner around the church entrance.

Houston's death marked the final chapter for the superstar whose fall from grace while shocking was years in the making. Houston had her first No. 1 hit by the time she was 22, followed by a flurry of No. 1 songs and multi-platinum records.

Over her career, she sold more than 50 million records in the United States alone. Her voice, an ideal blend of power, grace and beauty, made classics out of songs like "Saving All My Love For You," ''I Will Always Love You," ''The Greatest Love of All" and "I'm Every Woman." Her six Grammys were only a fraction of her many awards.

But amid the fame, a turbulent marriage to Brown and her addiction to drugs tarnished her image. She became a woman falling apart in front of the world.

Her last album, "I Look To You," debuted on the top of the charts when it was released in 2009 with strong sales, but didn't have the staying power of her previous records. A tour the next year was doomed by cancellations because of illness and sub-par performances.

Still, a comeback was ahead: She was to star in the remake of the movie "Sparkle" and was working on new music. Her family, friends and hard-core fans were hopeful.

The funeral is for invited guests only. Houston is to be buried next to her father, John Houston, in nearby Westfield, N.J.

New Jersey Education Consortium Hosts Live Chat with Space Station Astronauts

WASHINGTON ? Students and educators from Sussex County, N.J., will gather at Newton High School on Wednesday, Feb. 22, to further their space studies by speaking live with Expedition 30 Commander Dan Burbank and Flight Engineer Don Pettit aboard the International Space Station. The Earth-to-space communication will take place at 10 a.m. EST and be broadcast live on NASA Television.

The students participating in the event range from kindergarten through 12th grade. They represent the Newton, Andover Regional and Green Township school districts in Sussex County, all of which are part of the Tri-District Curriculum Consortium. U.S. Rep. Scott Garrett (R-NJ) is expected to join the group for this special event.

Reporters wanting to attend the event must contact Mary Decker at mdecker@newtonnj.org or 973-383-7392, ext. 230, by noon Tuesday, Feb. 21. Newton High School is located at 44 Reyerson Ave. in Newton.

The students participated in a variety of space-focused science, technology, engineering and mathematics (STEM) educational initiatives in preparation for the space station downlink. They also designed a special patch to commemorate their interaction with the orbiting astronauts.

This in-flight education downlink is one in a series with educational organizations in the United States and abroad to improve STEM teaching and learning. It is an integral component of NASA?s Teaching from Space Program, which promotes learning opportunities and builds partnerships with the education community using the unique environment of space and NASA?s human spaceflight program.

Also, if the temperature is cold you will require more calories and you?ll be burning more fat. You?ll also benefit from the thermogenic properties of protein.

Therefore if you would like protein and fat content on a hike, with very little waste packing (and therefore little extra poundage), then dry cured sausage such as chorizo is a productive means of carrying it. It is also pretty tasty.

You can eat it as it comes, without any cooking, with crackers or bread for instance. Or you can cook it: fry it in a pan (cut the sausage into 2-inch/4cm sections and slice down the middle or cut it into slices) or slice it relatively thin, skewer it on a (non toxic) green-wood stick and kebab it over your campfire (NB ensure the wood you are burning is also non-poisonous).

The cooked chorizo is delicious on its own cooked like this or you are able to add it to another dish such as a simple tomato-based pasta sauce. You can make a pasta sauce on the trail with a packet of powdered Bolognese sauce mixed with a powdered tomato soup. This could then be combined with your pasta. Straightforward, flavorsome, light, nutritive calorific and cheap.

For other options, just remember this: minmise the water content for the calories contained and ensure you have a good balance of carbohydrates, proteins and fats. And after you start seeing the possibility in familiar foods there?s masses of other foodstuff on the shelves of your area store that can be used in similar ways.
